Item Age And Growth Estimation Using Growth Band Counts Of Orange Mud Crab, Scylla Olivacea (Herbst, 1796) From Setiu Wetlands, Peninsular Malaysia(Universiti Malaysia Terengganu, 2018-05) Nurul Hasyima Binti IsmailOrange mud crab, Scylla olivacea (Herbst, 1796) is an important fishery products in Malaysia. Age study is important to generate the information on growth, recruitment, longevity, mortality for mud crab stock assessment. The aim of this study is to investigate the Growth Band Counts (GBC) deposited in thin sections of gastric mill of S. olivacea as age indicator for this species. A total of 90 wild caught S. olivacea were sampled from Setiu Wetlands, Peninsular Malaysia (February – October 2016) and 25 individuals of S. olivacea were reared in the hatchery of Institute of Tropical Aquaculture, Universiti Malaysia Terengganu for one year (March 2016 - March 2017). The periodicity of GBC in gastric mill of S. olivacea was validated using reared of known age samples. The results revealed that the presence of one GBC in reared S. olivacea were parallel to the age one year old of S. olivacea absolute age. One to three GBC were present in wild caught S. olivacea which indicated the age of one to three years old, respectively. There was a significantly positive relationship between Carapace Width (CW) and age, as the number of growth bands increased with putative age. The accuracy of bands counting was assessed by lower coefficient of variation values of reared and wild caught S. olivacea with 6.00 – 6.23% and 8.52 – 8.40%, respectively. Using the von Bertalanffy Growth Model equation, the growth parameters of S. olivacea was estimated and growth curves was fitted to CW at age data. The growth parameters such as asymptotic CW (CW∞), growth coefficient (K year -1 ) and initial condition parameter (t0) of the known age S. olivacea were narrower at 20.07 cm, 0.31 year -1 and - 0.20 than the wild samples at 28.01 cm, 0.36 year -1 and - 0.73 for female and at 29.34 cm, 0.38 year -1 and - 0.79 for male, respectively. The age composition suggested that S. olivacea population found in the Setiu Wetlands, Peninsular Malaysia has at least reached the age of 2 years old. Four diets containing different dosages of M. cajuputi leaves extracts; 0g/kg (C), 5g/kg (T1), 10g/kg (T2) and 15g/kg (T3) were added to the diet to investigate the effects of the respective treatments on the growth performance and blood parameter of Macrobrachium rosenbergii (mean weight of 3.17±0.46g). After the 45 days of feeding, prawns fed the diet containing 15g/kg of M. cajuputi leaves extracts significantly enhanced growth performance, which showed that weight gain (WG) and specific growth rate (SGR) was the highest amongst all treatments (P<0.05). Blood parameters (total hemocyte count (THC), differential hemocyte count (DHC), plasma glucose level, protein concentration and resistance against Aeromonas hydrophila) were examined 60 days post-feeding period. To evaluate its potential application in aquaculture, fertility and performance of desirable traits (e.g. growth, external morphology, flesh quality, disease resistance) were examined on reciprocal Fi and backcross hybrids. Reciprocal hybrids were partially fertile with various abnormalities in gonad development that resulted in low gamete production. Abnormalities observed in gonad development of all hybrid genotypes included a single ovary or testis, tumor-like and adipose-like tissues in ovaries, ovotestis, and retarded gonad. These losses will occur due to increasing ambient temperature and stagnation of transfer after being harvest. Quality deterioration will affect the consumer or market acceptences thus decrease the demand on the product. In this study, black tiger shrimp {Penaeus monodon) was obtained from a local supplier in Marang, Terengganu, killed and stored in an insulated box at 0°C for 9 days. For every storage period, the shrimp was processed prior to SDS-PAGE analysis and texture analysis.Item CHARACTERISATION OF THE Y-ORGAN IN THE ORANGE MUD CRAB, Scylla olivacea(Universiti Malaysia Terengganu, 2024-03) MUHAMMAD ACHDIATItem Characterization of bioflocculant-producing bacteria isolated from bioflocs of shrimp pond(Universiti Malaysia Terengganu, 2017-04) Nurul Fakriah Che HashimCharacterization of bioflocculant-producing bacteria isolated from bioflocs was prompted to explore marine bacteria that enhanced bioflocculation process in aquaculture wastewater treatment. Certain bacteria were believed to secrete extracellular polymeric substances (EPS) which response to the physiological stress encountered in the natural environment that can act as bioflocculants. The objectives of this study were identification of bioflocculant-producing bacteria isolated from bioflocs, evaluation on their flocculating activity and characterization of protein composition in extracellular polymeric substances (EPS) produced by identified bioflocculant-producing bacteria. In this study, standard methods on phenotypic and genotypic identification of bacteria including biochemical test and molecular approaches were employed. Six species known as Halomonas venusta, Bacillus cereus, Bacillus subtilis, Bacillus pumilus, Nitratireductor aquimarinus and Pseudoalteromonas sp. were successfully identified as bioflocculant-producing bacteria. Their flocculating activities were examined using Jar Test. The highest flocculating activity was exhibited by Bacillus cereus with 93% while iv Halomonas venusta showed the lowest record with 59% of flocculating activity. Extracted EPS that acted as bioflocculants were obtained from culture supernatant by using cold ethanol precipitation method. Protein content determination using Bradford assay indicated all six species of bioflocculant-producing bacteria showed different protein concentration that ranged between 1.377 μg/mL to 1.455 μg/mL. It is essential in aquaculture sector due to smaller size and good nutritional content suitable for the growth and survival of the fish larvae. It is also an important prey for other copepod and predators to mosquito larvae. The identification of the copepod species was done based on morphological characteristic only which is time consuming and need to be done on adult copepod only. Conservation measures which have been formulated and proposed have been translated into action by the appropriate federal and state agencies responsible for the conservation and management of sea turtles.Item Crossbreeding of Pangasius hypophtalmus and Pangasius nasutus and digestive system development of the hybrid larvae(Institut Akuakultur Tropika, 2008-04) Agus Putra, Abdul SamadA study to determine the feasibility of producing hybrid of Pangasius spp through crossbreeding between Pangasius hypophthalmus and Pangasius nasutus were conducted from June to August 2007. The result of the crossbreeding between P. hypophthalmus female and P. nasutus male (HN) showed a good breeding performance, where the average fertilization, hatching and deformation rates were 81.17%, 83.06% and 8.51%, respectively. However, crossbreeding between P. nasutus female and P. hypophthalmus male (NH) resulted in low breeding performance with the average fertilization, hatching and deformation rates at 60.96%, 54.61% and 14.68%, respectively. For the control, P. hypophthalmus (HH), the average fertilization, hatching and deformation rates were 93.18%, 89.73% and 4.14%, respectively; and for P. nasutus (NN), the average fertilization, hatching and deformation rates were 76.83%, 68.26% and 7.16%, respectively.
