Item A study on the genetic variation, cross compatibility and larval rearing of gouramis (trichogaster spp.)(Fakulti Agroteknologi dan Sains Makanan, 2003-03) Yik Sung, YeongGenetic variation of four species of gouramis from the genus Trichogaster namely T. trichoptems (three spot gourami), T. leeri (pearl gourami), T. pectoralis (snakeskin gourami) and T. microlepis (moonlight gourami) and three varieties of three spot gouramis namely yellow gourami, cosby gourami and lavender gourami were determined using Random Amplified Polymorphic DNA (RAPD) techniques. A total of seven individuals from each species and varieties were used for RAPD assay using two different primers namely OPA-11 and OPA-12. No polymorphism was detected in blue, lavender and cosby gouramis. The genetic distance between seven pearl gourami individuals were ranged from 0.0000 to 0.0370. The genetic distance obtained from moonlight gourami and snakeskin gourami ranged from 0.0000 to 0.4545 and 0.0000 to 0.2727, respectively.Item Study on genetic variability of oyster (Crassostrea iredalei, Faustino)in Peninsular Malaysia using RAPD-PCR Technique(Fakulti Agroteknologi dan Sains Makanan, 2003-09) Wan Bayani, Wan OmarThe random amplified polymorphic DNA (RAPD) technique was used to examine the genetic variability and relationship among individuals within and between populations of oysters (Crassostrea iredalei) from Terengganu, Kelantan, Perak and Kedah. The result of optimization of polymerase chain reaction (PCR) machine (Hybaid PCR Express, UK) showed that the best concentration of genomic DNA, magnesium chloride, dNTP- mixture, Fermentas Taq DNA Polymerase and primer for C. iredalei RAPD study were 50 ng, 3.0 mM, 0.4 mM, 2 U/|il and 0.4 |iM (10 picomoles) respectively. The machine produced clear RAPD banding patterns with an optimal annealing temperature of 36°C for 45 cycles. The genomic DNA was extracted from the oysters tissues by using phenol chloroform method. Twenty oligonucleotide primers (Kit A, Operon Technologies, Inc. Alameda, California) were screened and four primers were selected to amplify DNA from 80 samples of C. iredalei from the four populations. A total of 159 RAPD fragments (RAPDs) with 122 polymorphic fragments (76.73%) with the size ranging from 250 to 2000 base pairs (bp) were scored from the population.Item Storage stability of chilled minimally processed shredded cabbage(Universiti Putra Malaysia, 2003-09) Roshita IbrahimMinimally processed products are a rapidly growing segment for retail market and food service horticultural industry as they match the consumers' demand for product freshness and convenience. The shelf-life extension of these fresh-cut products is therefore relevant because of its economic impact. Packaging can be one of the important factors in determining the storage stability and quality of these freshcut products.Item Studies on cross-breeding of Clarias macrocephalus vs C.gariepinus and some aspects of their hybrid development(Fakulti Agroteknologi dan Sains Makanan, 2004-03) Masduki, Mohammad MorniA study on identifying the feasibility of producing hybrid catfish progeny through cross-breeding of Asian catfish (C. macrocephalus) and African catfish (C. gariepinus) were conducted at the Freshwater Hatchery of Kolej Universiti Sains dan Teknologi Malaysia. Results from three trials of reciprocal cross-breeding clearly showed that hybrid catfish progeny were successfully produced through cross-breed between C. macrocephalus female X C. gariepinus male (MG). This mating resulted in excellent breeding performances with fertilization, hatching and deformation rates averaged at 83.54%, 86.04% and 5.76%, respectively. However, cross-breed between C. gariepinus female X C. macrocephalus male (GM) resulted in low fertilization and hatching rates (48.90%, 40.35%).Item Growth and biochemical composition of sprulina platensis and s. fusiformis under fifferent culture conditions(Fakulti Agroteknologi dan Sains Makanan, 2004-06) Md. Rafiqul IslamA study on the growth and biochemical composition of Spirulina platensis and S. fusiformis were carried out at KUSTEM. Spirulina platensis was obtained from the Planktology Laboratory, KUSTEM, while S. fusiformis was obtained from the Marugappa Chettair Research Center (MCRC), Chennai, India. The culture was maintained by repeated transfer to liquid Zarouk medium. The effect of physico-chemical factors: nutrients, salinity, pH, temperature, light intensity and urea concentration on the growth and biochemical constituents of the two strains were investigated.Item Food shopping behaviour among muslim Malaysian students(University of Surrey, 2005) Siti Nurafifah, JaafarThe primary purpose of this study was to study the food shopping behaviour of Muslim students from Malaysia in University of Surrey. Thus, this study emphasises only four areas which are, (1) The difficulties encountered during shopping trips, (2) The difficulties experienced in adhering to Muslim diet, (3) The effect of acculturation to shopping behaviour and (4) The effect of acculturation on diet.Item Kajian mengenai pembiakan dan asuhan larva Latjanus johnii (Bloch)(Fakulti Agroteknologi dan Sains Makanan, 2005-01) Nik Razali, Nik LahKajian tentang pembiakan dan ternakan larva Lutjanus johnii (L. johnii) telah dilakukan di Pusat Pembiakan dan Penyelidikan, Tanjung Demong, Terengganu. Induk jantan dan betina L. johnii yang telah dipilih digunakan dalam kajian ini. Induk jantan yang mempunyai berat lebih 3.5 kg dan mempunyai sperma yang sedia untuk dilepaskan semasa pembiakan manakala induk betina juga mempunyai berat melebihi 3.5 kg dan mempunyai oosit yang bersaiz lebih 0.4 mm. Hempasan telur yang pertama berlaku 35 jam selepas disuntik. Induk jantan dan betina masing-masing disuntik dengan HCG pada dos 250 IU/kg dan 500 IU/kg. Lebih kurang 0.6 - 2.4 juta telur dikutip setiap kali pemungutan telur dilakukan dengan mencatat telur dihempaskan 0.3 x 106 hingga 0.5 x 106 telur/betina. Berdasarkan kepada data kajian yang telah dijalankan terdahulu didapati bahawa pembiakan L. johnii berlaku pada suku bulan pertama dan kedua setiap bulan.Item The performing of beef cattle integrated with oil palm plantations(Fakulti Agroteknologi dan Sains Makanan, 2005-08) Madzlan, Mohd SallehA study was conducted to determine the performance of beef cattle in oil palm plantation in three cattle zoning areas in Terengganu. Growth performance, conditioned age and management advantages of practices and nutrition were assessed. One hundred and ten heads of cattle of various age and sex were identified in each of the three selected farms and were allowed to graze freely under a controlled rotational grazing. Data on the initial and quarterly body weights were taken throughout the twelve months of the year of 2003. Fifteen random samples of the undergrowths using 1 m x 1 m quadrant were taken from each paddock for proximate composition of the feed.Item Design and development of a steady state close-cycle aquaculture system for intensive culture of fresh water fish species(Fakulti Agroteknologi dan Sains Makanan, 2006-01) Md AsaduzzamanThis study was to design and develop a steady-state conditioned closecycle aquaculture system to provide a conducive culture environment for intensive culture of fresh water fish species. Four major experiments were conducted to assess the efficiency of the system with varying ratios of volume of the fish rearing unit and volume of the biological filter (Aerobic) of 45:1, 23:1, 10:1 and 5:1 for the Experiment 1, Experiment 2, Experiment 3 and Experiment 4, respectively.Item Variability of greater yam (Dioscorea alata L.) cultivars in Malaysia(Fakulti Agroteknologi dan Sains Makanan, 2006-09) Andrew, NgadinDioscorea alata L. or greater yam is one of the old and highly variable tuber crop in Malaysia. Genetic variation of this species is poorly documented, which is becoming a hindrance in optimizing the use of its genetic resources available in this country. Hence, a study on the assessment of the morphological traits and molecular marker variations in the germplasms of D. alata collected in Malaysia was carried out in order to identify the most variables characters and to assess the genetic relationship among the variants.Item Fish and benthic communities within coral reef areas of Pulau Karah, Terengganu, Malaysia(Fakulti Agroteknologi dan Sains Makanan, 2006-10) Wan Mohd Rauhan, Wan HussinThe purpose of this study is to determine the abundance of fish and benthos, inside and outside coral areas in Pulau Karah, Terengganu. The abundance of the organisms was also compared between pre monsoon (September - October) and post monsoon (April - May). Seven stations were selected where Station 1 and 2 were inside coral area while Station 3 to 7 was outside coral area. The most dominant type of coral at Station 1 (10m depth) was found to be branching Acropora (ACB) while encrusting form of non-Acropora (CE) was the most dominant at Station 2 (15m depth). Station 1 has higher percentage of live coral (59.24%) compared to Station 2 with only 30.26% of live coral. Station 1 can be classified as in 'fair' condition and 'poor' condition was suitable to classify 'health' status of coral at Station 2.Item Phytochemical screening of Chromolaena odarata (L.) extracts for anti-bacterial, anti oxidant and anti-leukaemic properties(Fakulti Agroteknologi dan Sains Makanan, 2006-10) Nurul Huda, Abd Kadir @ Abdul RahmanChromolaena odorata (L.) is a noxious weed that can be found spreading widely in tropical countries. Traditionally, this plant was used to cure wounds and minor burns. The polyphenolic compounds in the crude extract and terpenoids compounds in the essential oil of Chromolaena odorata were found to be responsible in inhibiting bacterial and HL 60 cells (Human Promyelocytic Leukaemia) growth. In addition, it also showed the potential as anti-oxidant agent. The content of Chromolaena odorata crude extract that contain polyphenol compounds was determined using LC-MS, whereas the compositions of the essential oil that contain terpenoid compounds were obtained using GC-MS.Item Effect of seeed cluster and gibberellic acid (GA3)on the gender development of salak (Salacca edulis reinw)(Fakulti Agroteknologi dan Sains Makanan, 2007-09) Razifah, Mohd RazaliA series of experiments on growth and development of Salacca edulis REINW. were conducted in the greenhouse of Universiti Malaysia Terengganu (UMT) and in the field on undeveloped agriculture land (bris soil) in Tumpat, Kelantan. The nursery study was undertaken for six months followed by a field study for 24 months. In the nursery study, seeds were soaked in 0, 10, 50 and 100 ppm gibberellic acid (GA3) and planted in individual polybags according to their type of seed clusters which comprised one, two and three seeds per fruit. The polybags were placed under warm, moist and shady conditions to avoid stress due to dryness and over-watering. The seedlings were then left for six months in the greenhouse before they were transplanted into the field.Item Kajian in vitra jangkitan fluk darah sanguinicola armata plehn, 1905 (Digenea: Sanguinicolidae) pada siput gyraulus convexiusculus dan ikan kap kepala besar aristichthys nobilis(Fakulti Agroteknologi dan Sains Makanan, 2007-09) Nor Asilah, SemanBeberapa aspek jangkitan fluk darah Sanguinicola armata terhadap siput Gyraulus convexiusculus dan ikan kap kepala besar Aristichthys nobilis telah dijalankan. Di dalam kajian takungan jangkitan A. nobilis yang dijangkiti S. armata telah didedahkan kepada siput G. convexiusculus dan juvenil A. nobilis yang bebas daripada jangkitan. Kajian ini dijalankan bagi mengatasi masalah untuk mendapatkan sumber S. armata. Keputusan menunjukkan bahawa terdapat saling jangkitan di antara semua perumah terhadap S. armata.Item Some aspects of biology and population dynamics of the goby pseudapocryptes elongatus (Cuvier 1816) in the mekong delta(Fakulti Agroteknologi dan Sains Makanan, 2008-01) Tran, Dac DinhThe goby, Pseudapocryptes elongatus (Cuvier, 1816), is a common and commercial fish in the coastal areas of the Mekong Delta, Vietnam. This species is recently cultured in semi-intensive and intensive farming system. However, the seeds are collected only from the wild and not enough for aquaculture. Meanwhile, studies on the biology of this species are still limited, especially on the reproductive biological characteristics. Therefore, some aspects of the biology and population dynamics of the goby Pseudapocryptes elongatus distributed in the coastal mud flat areas of the Mekong Delta were investigated from January 2004 to June 2005. The fish populations were collected and examined to determine the reproductive biological characteristics. Threats to its natural population were identified and the results can be used for sustainable use of the fish populations and aquaculture purposes.Item Conventional and molecular characterization of Vibro parahaemolyticus, vibrio alginolyticus and vibro vulnificus in oyster, crassostrea iredalei from east coast , Malaysia(Fakulti Agroteknologi dan Sains Makanan, 2008-04) Ruhil Hayati, HamdanOyster is a filter feeder that consumes a large volume of bacteria during ingestion activity. Totally, 28, 10 and 15 isolates of V. parahaemolyticus, V. vulnificus and V. alginolyticus were isolated from oyster, respectively. The morphological, physiological and biochemical studies were done with addition of commercial identification system BBL Crystal™ Kit. All isolates were motile Gram-negative; straight and curved rod; non-luminous, oxidase and catalase positive; sensitive to 0/129 (150(ig), glucose fermentative, indole formation, able to utilize starch, positive for both Oxidative and Fermentative, and p-hemolytic. All isolates did not produce H2S. All isolates also showed negative reactions for Voges Proskauer, ONPG and tetrazoliumItem Keperluan protein dalam diet untuk tumbesaran dan kematangan induk kelah, Tor tambroides (bleeker)(Fakulti Agroteknologi dan Sains Makanan, 2008-11) Suhaini, Ahmat @ AmiruddinPemakanan merupakan elemen penting dalam mengekalkan tahap kesihatan, tumbesaran dan kematangan bagi spesies ikan yang dikultur. Protein diperlukan untuk membekalkan tenaga dalam badan dan diperlukan semasa proses sintesis hormon, antibodi, enzim, dan pembentukan tisu. Kajian ini telah dijalankan untuk mengetahui keperluan protein dalam diet terhadap tumbesaran dan kematangan induk kelah, Tor tambroides. Dalam kajian ini, 4 diet yang berbeza telah diberikan kepada induk T. tambroides yang mengandungi paras protein sebanyak 30%, 35%, 45% dan 55%. Di sepanjang 120 hari tempoh kajian, induk diberi diet tersebut pada kadar 3% daripada berat badan. Kajian ini telah dijalankan di hatcheri Universiti Malaysia Terengganu.Item Physiological and growth responses of pineapple (Ananas comosus (L.) merrill) in different level of salinity and drought stress under in vitro culture(Fakulti Agroteknologi dan Sains Makanan, 2009-02) Nur Suraya, AbdullahThe areas along the coastal line of the East Coast of Peninsular Malaysia are largely covered with the 'Beach Ridges Interspersed with Swales' (BR1S) soil, where it is commonly faced with salt deposit problem and having severe drought condition during the middle of the year. The BR1S soil contains low water holding capacity, less nutrient and not suitable for cultivation of many crops species. Pineapple {Ananas comosus (L.) Merrill) is recorded as a crop originated from the arid region of the Central America, and it is believed to be tolerant under drought condition and might be very suitable to be cultivated in the BR1S (Beach Ridges Interspersed with Swales) soil area. Thus, a study was conducted to examine the effect of salinity and drought stress on three cultivated varieties (cultivar) of pineapple, namely "N36, Morris and Sarawak, by using the tissue culture techniques with the aim at determining the best pineapple variety for and cultivating under the BR1S soil condition.Item Characteristation of Edwardsiella tarda isolated from local Calrias gariepinus and potential remedies from edible plants(Fakulti Agroteknologi dan Sains Makanan, 2009-05) Lee Seong WeiThis study characterised 7 isolates of Edwardsiella tarda from African catfish, Clarias gariepinus. The isolated E. tarda were identified using a combination of 29 conventional tests and commercial kit. Random amplification of polymorphic DNApolymerase chain reaction (RAPD PCR) analysis was done on these bacterial isolates using three universal primers, namely WTP, (GTG)s and M13 universal. Characterisation of whole cell protein profiles of the isolates were also carried out using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). In the present study, minimum inhibitory concentration (MIC) values of 20 edible plant extracts against E. tarda were determined through a two fold microdilution technique.Item Identification, prevalence, mean intensity and abundance of trichodinids (protozoa: ciliophora: peritrichida) from cyprinus carpio (KOI) at Sungai Siput, Perak(Fakulti Agroteknologi dan Sains Makanan, 2009-06) Syaidatunorhani Abdul MananTrichodinid parasites belonging to the order Mobilina and family Trichodinidae are found infecting the skin, fins and gills of Cyprimis carpio (Koi) of Sungai Siput, Perak. Studies were carried out at 2 stations, namely Sl-Rimba Panjang and S2-Sungai Buloh starting from April 2006 to June 2007. Diagnosis of 120 Koi fish revealed the presence of six species and one genus of Trichodinid ciliates, namely Trichodina acuta, Trichodina heterodentata, Trichodina nobilis, Trichodina mutabilis, Trichodina nigra, Trichodinella epizootica and Tripartiella sp.
